Materials

M Tris-HCl (pH 6.8)
Sodium dodecyl sulfate (SDS)
glycerol (87%)
M EDTA (pH 8.0)
Bromophenol blue

Experiment settings

- Volume SDS-sample buffer	ml
- Concentration fold:	X

Step 1

Prepare ml of SDS-sample buffer X by adding:

ml Tris-HCl (pH 6.8)
g SDS
ml glycerol (87%)
ml EDTA (pH8.0)
mg bromophenol blue
ml ß-mercaptoethanol

Step 2

Add ml H₂O to make a total volume of ml and mix well

Step 3

Make aliquots and store at -20ºC

DTT may be added instead of ß mercaptoethanol as a reducing agent.

Thsi recipe contains EDTA. EDTA is not really required as I see from other protocols. Is it just to chelate divalent and trivalent cations in solution? Is this really important?
Also, adding 1 mL of 0,5 M EDTA at pH 8,0 to 1mL of 1M Tris pH 6,8 resu

SDS sample buffer without EDTA also works fine! Just copy this protocol and change it if you like.
